It is the biggest step there is. A single pane is one thin sheet between you and the weather. A modern sealed unit has two panes, a Low-E coating, argon between them and a warm-edge spacer, so the room holds its heat far longer and the draught round the sash stops as well.
Heat leaves a house through whatever is coldest, and old glazing with tired seals is usually near the top of that list. Energy rated windows slow that down, which matters most in a house that is costly to heat in the first place. The surveyor will point out which openings are losing the most.
Yes, usually a lot. Moisture lands on the coldest surface in the room, and a warm inner pane gives it far less reason to settle there. Trickle vents, where the regulations call for them, let damp air out of the room instead of trapping it against the glass and the sill.
